That should work. Simpler is the use of a mirror which reflects the
light of the scanner to the back of the transparency. Such mirrors are
included in some HP flatbed scanners and work with other brands too.
I am using this setup for low resolution scans.

However, flatbed scanners don't deliver the same image quality 
as dedicated film scanners. You will get better resolution and
depth using the method I mentioned earlier.
